Description
Tramadol hydrochloride is a centrally-acting synthetic opioid analgesic binding to specific opioid receptors. It is a non-selective, pure agonist at mu, delta and kappa opioid receptors with a higher affinity for the mu receptor. Other mechanisms, which may contribute to its analgesic effect, are inhibition of neuronal re-uptake of noradrenaline and enhancement of serotonin release.
Tramadol hydrochloride does not promote histamine release.

Indications & Dosage
Indications:
Tramadol injection (VESNON-V100) is indicated for the treatment of moderate to severe pain.
Dosage and Administration
A dose of 50 to 100 mg may be given every 4 to 6 hours by intramuscular injection, intravenous injection over 2 to 3 minutes, or by intravenous infusion. For tire treatment of postoperative pain, the initial dose is 100 mg followed by 50mg every 10 to 20 minutes if necessary to a total maximum (including the initial dose) of 250 mg in the first hour. Thereafter, doses are 50 to 100 mg every 4 to 6 hours up to a total daily dose of 600mg.
Elderly:
The usual doses maybe used. However, it is reported that volunteers aged over 75 years, the elimination half-life of tramadol was increased by 17% following oral administration.
Renal impairment /Hepatic impairment:
The dosage interval should be increased to 12 hours in patients with renal or liver impairment. Tramadol should not be used if there is severe renal impairement. Removal by haemodialysis is reported to be very slow.
